Immortalization Method	Transfection with the retroviral vector (pLXSN–hTERT)
Description	Primary oral epithelial cells were derived from patients undergoing removal of impacted teeth and were immortalized with the retroviral transfection of the pLXSN-hTERT vector to establish the Immortalized Human Oral Mucosal Epithelial Cells (hTERT-OME). The cells are also considered to be oral keratinocytes and have maintained their stem cell properties. hTERT-OME cells are a valuable tool for research in the biology of oral epithelial cells.
Procedure Overview
Propagation	Use of PriCoat™ T25 Flasks (G299) or Applied Cell Extracellular Matrix (G422) is required for cell adhesion to the culture vessels. Grow cells in ECM-coated culture vessels with the following conditions. The base medium for this cell line is Prigrow IV medium available from abm (TM004). To make the completed growth medium, add the following components to the base medium: fetal bovine serum (TM999) to final concentration of 10%, 200 µg/ml G418 (G271), and 1% Penicillin/Streptomycin (G255). Atmosphere: air: 95%, CO₂: 5%; Temperature: 37.0°C.
